I don't understand the question what are we comparing the char of letters to in the for loop?

The question is not clear enough...

ScrabblePlayer.java
public class ScrabblePlayer {
  private String mHand;
  private int count;

  public ScrabblePlayer() {
    mHand = "";
  }

  public int getTileCount(){
    for (char letter: mHand.toCharArray()){
      if ( letter ...... ){
        count++;
      }
    } return count;
  }

  public String getHand() {
   return mHand;
  }

  public void addTile(char tile) {
    // Adds the tile to the hand of the player
    mHand += tile;
  }

  public boolean hasTile(char tile) {
   return mHand.indexOf(tile) > -1;
  }
}

You're supposed to be comparing it to a char that you accept as a parameter in the getTileCount method.

public int getTileCount(char match){
  // for each char
  // if char == match
  // count++
}
